초록(영문) | The aim of this study was to evaluate the effects of collagen and mussel adhesive protein (MAP) coating on endothelial cell adhesion under perfusion culture conditions. Three kinds of scaffolds were prepared: (I) PCL scaffold fabricated by salt leaching method, (2) PCL scaffold coated with collagen, (3) PCL scaffold coated with collagen and MAP. Scaffolds were loaded in PDMS chambers which were connected with perfusion system. The magnitude of shear stress acting on the scaffold surface was estimated by FEM analysis. After seeding cells on the scaffold surface, they were cultured for 6 days under perfusion condition. DNA quantification, real-time PCR, SEM imaging and immnostaining were used for evaluating the cell adhesion. The results showed that perfusion culture could cause cell detaclnnent at the initial state, but it promoted gene expressions related with cell adhesion and ECM secretion in the long term. Especially, MAP coating and perfusion condition showed synergetic effects on gene expressions and ECM secretion. |
